Will,

Since speakup is a kernel based driver, speakup will work with 
any x86 linux that uses the standard linux kernel. If you can build 
a standard kernel for puppy linux then you can build a modified 
kernel with speakup support. 

Having said that, puppy linux is a small live CD based distro. 
To make it work you would have to get the source, patch the
kernel for speakup, recreate the CD ISO image and burn it.
